The short version of peptide purity fits in a sentence. The long version — which is the one that helps — is below.
Reviewed 2026-03-18. Anything still debated is marked as such rather than presented as settled.
Because the substance circulates mainly through informal markets, verification is a recurring theme in technical discussion. Independent analyses have found that labeled content and actual content can diverge, and that purity varies between samples. Analytical laboratories use reversed-phase chromatography to separate components and mass spectrometry to confirm identity. Isotope-labeled internal standards improve quantification in complex matrices. Such methods describe what a sample contains but say nothing about its sterility, lawful status, or suitability for any use. Open questions remain about how consistently testing is applied across the supply chain.
Regulatory treatment of this peptide varies by country. It holds no marketing authorization as a medicine in the United States, the European Union, or most other jurisdictions. Some countries classify products containing it as prescription-only or unlicensed medicines, which restricts lawful supply. Authorities have issued public notices warning that unregulated products may contain undeclared or incorrect ingredients. The molecule also appears on prohibited lists for competitive sport. These measures address supply oversight rather than any approved therapeutic role.
Human data remain limited and mostly short-term. Reports describe small trials and observational accounts rather than large controlled studies, so questions about dose-response relationships and long-term effects on melanocytes stay open. Whether repeated exposure alters naevus behaviour is not settled in the published record. Researchers also note that self-administered use outside clinical settings makes actual exposure difficult to quantify. Statements about efficacy and safety should therefore be read as preliminary rather than established.
Melanotan II is a synthetic cyclic heptapeptide that acts as an agonist at melanocortin receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide involved in pigment production. The analogue carries a lactam bridge that constrains the ring and slows enzymatic breakdown relative to the native hormone. In research literature it appears under several abbreviations, and naming conventions are not fully standardized. Published descriptions usually place it within the broader melanocortin agonist family.

Independent verification is central to quality control because the compound is not produced under pharmaceutical manufacturing standards. Third-party laboratories can measure purity, identity, residual solvents, and microbial contamination, though the scope of testing varies between services. Reported analyses of vendor samples have shown batch-to-batch variation in peptide content and the presence of truncated or oxidized species. How much of this variation reflects synthesis conditions versus storage and shipping is not well characterized. No harmonized reference standard exists for the material as sold.
Handling guidance for melanotan II follows general practice for small synthetic peptides rather than a product-specific monograph. Lyophilized powder is typically kept at minus twenty degrees Celsius or colder, protected from light and moisture, because warmth and humidity accelerate degradation. Once reconstituted, solutions are usually refrigerated and used within a short window, as hydrolysis and microbial growth both become concerns. Repeated freeze-thaw cycles are generally avoided. These conventions come from laboratory peptide chemistry and not from formal stability studies on this specific compound.
Analytical confirmation of identity relies on mass spectrometry, most often coupled to liquid chromatography. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and provides a purity estimate based on peak area. Electrospray ionization mass spectrometry then confirms the expected molecular mass, while tandem mass spectrometry can map the fragment sequence. For research-grade material, these two techniques together form the standard minimum. Purity figures reported by vendors are frequently not traceable to an independent laboratory.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Regulatory status differs by country, and in many places supplying the compound for human consumption is unlawful. Vendors frequently label material as intended for research use only, a designation that shifts stated purpose but does not create a legal pathway for personal use. Certificates of analysis accompanying such products vary widely in detail and provenance. Third-party testing exists but is voluntary, and results are rarely linked to a specific lot in a publicly verifiable way.
Identity testing for a synthetic peptide relies on several complementary methods. Reversed-phase high-performance liquid chromatography separates the target from related impurities and reports purity as a percentage of total peak area. Mass spectrometry confirms molecular mass and can reveal deletions or truncations. Amino acid analysis and peptide mapping provide sequence-level confirmation, while counter-ion content and residual solvents are measured separately. A purity figure alone does not establish identity, so a complete dataset combines chromatographic and spectrometric evidence.
Lyophilized peptide powder is generally stored frozen, protected from light and moisture. Tryptophan residues are susceptible to oxidation, and the lactam bridge can hydrolyze under strongly acidic or basic conditions. Solutions prepared for laboratory work degrade faster than dry powder, and repeated freeze-thaw cycles accelerate loss. Common practice is to aliquot solutions before freezing and to avoid alkaline buffers. Reported stability windows vary with concentration, buffer, and temperature, so exact shelf lives are method-specific rather than universal.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
=== New owner === On 23 February 2023, Giant's operator Dairy Farm announced that it was exiting the grocery retail market in Malaysia, citing increasing competition. Dairy Farm's grocery business in Malaysia was sold to Macrovalue Sdn Bhd led by Andrew Lim Tatt Keong, a local businessman and entrepreneur. Dairy Farm, however, would still remain the operator of the Cold Storage and Giant franchise in Singapore until 2025, where it will be sold to Macrovalue Pte Ltd in March 2025. Following the acquisition, Macrovalue announced that Giant Hypermarket in both Singapore and Malaysia will be rebranded as Giant Mall as the parent company attempts to attract more foot traffic and cut down losses.
=== Foreign policy === Gallrein supported the 2026 U.S. intervention in Venezuela. Commenting on the 2026 Iran War, Gallrein said on X on February 28, 2026, that the United States' attack on Iran: "sends a clear message to the world: The United States will not sit back while a hostile regime continually threatens our Nation, our allies, and global stability as Iran has for decades as the leading state sponsor of terrorism been responsible for the death of U.S. citizens and servicemembers.... Decisive action, clear objectives, and overwhelming strength save American lives and prevent larger wars, death, and destruction."
==== Anti-inflammatory treatments ==== Topical corticosteroids are effective in short-term treatment of seborrhoeic dermatitis and are as effective or more effective than antifungal treatment with azoles. These are sometimes used for a few weeks at a time. There is also evidence for the effectiveness of topical calcineurin inhibitors like tacrolimus and pimecrolimus as well as lithium salt therapy. Calcineurin inhibitors were also effective in reducing the growth of Malassezia, offering two routes by which they may treat seborrhoeic dermatitis. Medications such as calcineurin inhibitors are not often used in individuals with seborrhoeic dermatitis who are immunocompromised because they cause further immune suppression. Oral immunosuppressive treatment, such as with prednisone, has been used in short courses for seborrhoeic dermatitis, as a last resort due to its potential side effects.
"In 1990 members of the Bush administration ... cited Security Council resolutions as sufficient support for the President to act militarily against Iraq without congressional authority." A series of UN Security Council resolutions and Arab League resolutions were passed regarding Iraq's invasion of Kuwait. Resolution 678, passed on 29 November 1990, gave Iraq a withdrawal deadline until 15 January 1991 and authorized "all necessary means to uphold and implement Resolution 660", and a diplomatic formulation authorizing the use of force if Iraq failed to comply. To ensure the US received economic backing, James Baker went on an 11-day journey to nine countries in September 1990, which the press dubbed "The Tin Cup Trip". The first stop was Saudi Arabia, which had already granted permission to the US to use its facilities. Baker believed that Saudi Arabia should assume some of the cost of to defend it. When Baker asked King Fahd for $15 billion, the King agreed, with the promise that Baker ask Kuwait for the same amount. The next day, 7 September, Baker did just that, and the emir of Kuwait, displaced in a Sheraton hotel outside Kuwait, agreed. Baker moved to enter talks with Egypt, whose leadership he considered "the moderate voice of the Middle East". President Mubarak was furious with Saddam for his invasion, and that Saddam had assured Mubarak that an invasion was not his intention. Egypt received approximately $7 billion in debt forgiveness for providing support and troops for the US-led intervention.
== Historical significance == Enteropeptidase was discovered by Ivan Pavlov, who was awarded the 1904 Nobel Prize in Physiology or Medicine for his studies of gastrointestinal physiology. It is the first known enzyme to activate other enzymes, and it remains a remarkable example of how serine proteases have been crafted to regulate metabolic pathways. The inert function of digestive enzymes within the pancreas was known, as compared to their potent activity within the intestine, but the basis of this difference was unknown. In 1899, Pavlov's student, N. P. Schepowalnikov, demonstrated that canine duodenal secretions dramatically stimulated the digestive activity of pancreatic enzymes, especially trypsinogen. The active principle was recognized as a special enzyme in the intestine that could activate other enzymes. Pavlov named it enterokinase. The debate of whether enterokinase was a cofactor or enzyme was resolved by Kunitz, who showed that the activation of trypsinogen by enterokinase was catalytic. In the 1950s, cattle trypsinogen was shown to be activated autocatalytically by cleavage of an N-terminal hexapeptide. The more precise IUBMB name enteropeptidase has been in existence since 1970. However, the original name ‘enterokinase’ has a long history and remains in common use.

== Economic impact == Though the many advances of HPLC and monoliths are highly visible within the confines of the analytical and pharmaceutical industries, it is unlikely that general society is aware of these developments. Currently, consumers may witness technology developments in the analytical sciences industry in the form of a broader array of available pharmaceutical products of higher purity, advanced forensic testing in criminal trials, better environmental monitoring, and faster returns on medical tests. In the future, presumably, this may not be the case. As medicine becomes more individualized over time, consumer awareness that something is improving their quality of care seems more likely. The further thought that monoliths or HPLC are involved is unlikely to concern the general public, however. There are two main cost drivers behind technological change in this industry. Though many different analytical areas use LC, including food and beverage industries, forensics labs, and clinical testing facilities, the largest impetus toward technology developments comes from the research and development and production arms of the pharmaceutical industry. The areas in which high-throughput monolithic column technologies are likely to have the largest economic impact are R&D and downstream processing. From the Research and Development field comes the desire for more resolved, faster separations from smaller sample quantities. The only phase of drug development under direct control of a pharmaceutical company is the R&D stage.
=== Amino acid enrichment for certain functional categories of LCRs === A bioinformatics analysis of prokaryotic LCRs identified 5 types of amino acid enrichment, for certain functional categories of LCRs:
==== Advantage of argon ==== Making the plasma from argon, instead of other gases, has several advantages. First, argon is abundant (in the atmosphere, as a result of the radioactive decay of potassium) and therefore cheaper than other noble gases. Argon also has a higher first ionization potential than all other elements except He, F, and Ne. Because of this high ionization energy, the reaction (Ar+ + e− → Ar) is more energetically favorable than the reaction (M+ + e− → M). This ensures that the sample remains ionized (as M+) so that the mass spectrometer can detect it. Argon can be purchased for use with the ICP-MS in either a refrigerated liquid or a gas form. It should have a guaranteed purity of 99.9% Argon at a minimum. It is important to determine which type of argon will be best suited for the specific situation. Liquid argon is typically cheaper and can be stored in a greater quantity as opposed to the gas form, which is more expensive and takes up more tank space. If the instrument is in an environment where it gets infrequent use, then buying argon in the gas state will be most appropriate as it will be more than enough to suit smaller run times and gas in the cylinder will remain stable for longer periods of time, whereas liquid argon will suffer loss to the environment due to venting of the tank when stored over extended time frames. However, if the ICP-MS is to be used routinely and is on and running for eight or more hours each day for several days a week, then going with liquid argon will be the most suitable.
Bottle fermentation Some beers undergo an additional fermentation in the bottle giving natural carbonation. This may be a second and/or third fermentation. They are bottled with a viable yeast population in suspension. If there is no residual fermentable sugar left, sugar or wort or both may be added in a process known as priming. The resulting fermentation generates CO2 that is trapped in the bottle, remaining in solution and providing natural carbonation. Bottle-conditioned beers may be either filled unfiltered direct from the fermentation or conditioning tank, or filtered and then reseeded with yeast.
At the Potsdam Conference starting in late July 1945, the Allies met to decide how to administer the defeated Nazi Germany, which had agreed to unconditional surrender nine weeks earlier on May 7 and May 8, 1945, VE Day. Serious differences emerged over the future development of Germany and Eastern Europe. Administration officials favoring cooperation with the Soviet Union and the incorporation of socialist economies into a world trade system were marginalized. The UK was represented by a new prime minister, Clement Attlee, who had replaced Churchill after the Labour Party's defeat of the Conservatives in the 1945 general election. The US had invited Britain into its atomic bomb project but kept it secret from the Soviet Union. Stalin became aware that the Americans were working on the atomic bomb through his spy network, however. One week after the end of the Potsdam Conference, the US bombed Hiroshima and Nagasaki. Shortly after the attacks, Stalin protested to US officials when Truman offered the Soviets little real influence in occupied Japan. Stalin was also outraged by the actual dropping of the bombs, calling them a “superbarbarity” and claiming that “the balance has been destroyed ... That cannot be”. The Truman administration intended to use its ongoing nuclear weapons program to pressure the Soviet Union in international relations. The immediate end of war material shipments from America to the USSR after the surrender of Germany also upset some politicians in Moscow, who believed this showed the US had no intentions to support the USSR any more than they had to.

Two policemen disguised as orienteers, Bertil Brosved and Ulf Högenberg, tried to arrest him at the same time as Olofsson pulled a pistol out of his waistband and fired two shots. Högenberg was hit in the shoulder. Olofsson was first sentenced to ten years in prison, but the Court of Appeal changed the sentence to eight years. Norgren was sentenced to 12 years, which was the most severe punishment a Swedish court could impose at that time. On 4 February 1969, Olofsson escaped from Kumla Prison and fled to the Canary Islands. He then entered West Germany on a fake passport, flying to Frankfurt am Main, where he lived until he was arrested by the German police. He was escorted to the ferry in Travemünde where two Swedish police officers met him. He was then taken via Malmö back to the Kumla Prison. Two months before he was to be released, he escaped again, from Lingatan Prison, an open institution in Bohuslän. On 2 February 1973, he was arrested in the dining room of the Kurhotel in Ulricehamn. Police had received a tip from a cleaning lady who had seen a gun in his hotel room. At the time of his arrest, he had been on the run for seven months and had robbed a bank in Gothenburg. In May 1973, he was sentenced to six years in prison and transported to the Kalmar Prison.
Scar free healing is the process by which significant injuries can heal without permanent damage to the tissue the injury has affected. In most healing, scars form due to the fibrosis and wound contraction, however in scar free healing, tissue is completely regenerated. During the 1990s, published research on the subject increased; it is a relatively recent term in the literature. Scar free healing occurs in foetal life but the ability progressively diminishes into adulthood. In other animals such as amphibians, however, tissue regeneration occurs, for example as skin regeneration in the adult axolotl.
== Adverse effects and precautions == Generally, oral CoQ10 supplementation is well tolerated. The most common side effects are gastrointestinal symptoms (nausea, vomiting, appetite suppression, and abdominal pain), rashes, and headaches. Some adverse effects, largely gastrointestinal, are reported with intakes. Doses of 100–300 mg per day may induce insomnia or elevate liver enzymes. The observed safe level risk assessment method indicated that the evidence of safety is acceptable at intakes up to 1200 mg per day. Caution should be observed in the use of CoQ10 supplementation in people with bile duct obstruction and during pregnancy or breastfeeding.
=== Substrate-like inhibitors === Substrate-like inhibitors (Figure 4) are more common than the non-substrate-likes. They bind either covalently or non-covalently and have a basic structure where the P1-substituent occupies the S1-pocket and the P2-substituent occupies the S2-pocket. Usually they contain a proline mimetic that occupies the S1-pocket. Large substituents on the 2-cyanopyrrolidine ring are normally not tolerated since the S1-pocket is quite small. Since DPP-4 is identical with the T-cell activation marker CD26 and DPP-4 inhibitors are known to inhibit T-cell proliferation, these compounds were initially thought to be potential immunomodulators. When the function against type 2 diabetes was discovered, the cyanopyrrolidines became a highly popular research material. A little later vildagliptin and saxagliptin, which are the most developed cyanopyrrolidine DPP-4 inhibitors to date, were discovered.

Only a small number of early-stage human studies have been reported, and most were limited in size and duration. No large late-stage program has established a general efficacy or safety profile. The evidence base is therefore thin compared with approved medicines.
The peptide is listed as prohibited in competitive sport, so laboratories develop detection methods and monitor for its presence. Its appearance in enforcement reports reflects that classification rather than any approved athletic use. Analytical work in this area focuses on identifying the molecule and its breakdown products in biological samples.
Chromatographic separation combined with mass spectrometry identifies the peptide and estimates its amount in a sample. Testing can confirm whether the expected molecule is present and at what concentration. It cannot establish how a product was manufactured or whether it meets any purity standard.
It is a synthetic cyclic heptapeptide and a non-selective melanocortin receptor agonist. Structurally it is modelled on alpha-melanocyte-stimulating hormone, a peptide that occurs naturally in the body.
