Melanotan-2 is a synthetic linear peptide built from seven amino acids arranged in a short chain. Its sequence is commonly written as Ac-Nle-Asp-His-D-Phe-Arg-Trp-Lys-NH2, which includes a modified N-terminus and an amidated C-terminus. The molecule belongs to the melanocortin family and acts as a receptor agonist. Structural features such as the D-phenylalanine residue and the Nle substitution are associated with increased stability against enzymatic degradation relative to the natural parent peptide.
The compound emerged from research programs in the 1980s that examined analogues of alpha-melanocyte-stimulating hormone for pigmentation and photoprotection. Investigators modified the native sequence to extend activity duration and potency. A related analogue, afamelanotide, was developed within the same broad line of inquiry and eventually gained approval in certain jurisdictions for a rare light-sensitivity condition. Melanotan-2 itself did not progress through the same regulatory route and has no approved therapeutic indication.
Melanocytes are the pigment-producing cells of the skin, and they carry melanocortin-1 receptors on their surface. When the receptor is activated, cyclic adenosine monophosphate rises inside the cell and raises the activity of enzymes such as tyrosinase, which increases melanin output. Melanotan-2 binds melanocortin-1 receptors in vitro and in animal models, and this binding is generally described as the basis for the tanning effect. Other receptors account for different effects: melanocortin-4 receptors contribute to appetite and erectile signalling, while melanocortin-3 and melanocortin-5 receptors contribute to energy balance and exocrine function.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.

== Quotes == "The First Cell arose in the previously prebiotic world with the coming together of several entities that gave a single vesicle the unique chance to carry out three essential and quite different life processes. These were: (a) to copy informational macromolecules, (b) to carry out specific catalytic functions, and (c) to couple energy from the environment into usable chemical forms. These would foster subsequent cellular evolution and metabolism. Each of these three essential processes probably originated and was lost many times prior to The First Cell, but only when these three occurred together was life jump-started and Darwinian evolution of organisms began." (Koch and Silver, 2005) "The evolution of modern cells is arguably the most challenging and important problem the field of Biology has ever faced. In Darwin's day the problem could hardly be imagined. For much of the 20th century it was intractable. In any case, the problem lay buried in the catch-all rubric "origin of life"--where, because it is a biological not a (bio)chemical problem, it was effectively ignored. Scientific interest in cellular evolution started to pick up once the universal phylogenetic tree, the framework within which the problem had to be addressed, was determined. But it was not until microbial genomics arrived on the scene that biologists could actually do much about the problem of cellular evolution." (Carl Woese, 2002)
==== Partition of Vietnam ==== The Geneva Conference recognized the 17th parallel north as a "provisional military demarcation line", temporarily dividing the country into two zones. Operation Passage to Freedom began in August, consisting of the evacuation of Vietnamese civilians from communist North Vietnam to pro-Western South Vietnam. The Geneva Accords promised elections in 1956 to determine a national government for a united Vietnam. Neither the United States government nor the State of Vietnam signed the 1954 Geneva Accords. With respect to the question of reunification, the non-communist Vietnamese delegation objected strenuously to any division of Vietnam, but lost out when the French accepted the proposal of Viet Minh delegate Phạm Văn Đồng, who proposed that Vietnam eventually be united by elections under the supervision of "local commissions". The United States countered with what became known as the "American Plan", with the support of South Vietnam and the United Kingdom. The American Plan provided for unification elections under the supervision of the United Nations, but was rejected by the Soviet delegation. From his home in France, Bảo Đại appointed Ngo Dinh Diem as Prime Minister of the State of Vietnam. With American support, in 1955 Diem used a referendum to remove the former Emperor and declare himself the president of the State of Vietnam. When the elections failed to occur, Viet Minh forces that had remained in South Vietnam were activated and began to fight the government.
=== 2nd generation cephalosporins === Early second generation cephalosporins are very similar in basic structure to the first generation. Loracarbef however does not have the normal dihydrothiazine ring but is a carbacephem that has a carbon atom in the ring instead of a sulfur atom making it a tetrahydropyridine ring. This chemical property gives loracarbef better stability in plasma while retaining oral absorption characteristics and affinity for binding to PBP. The 7-phenyl-glycine makes it orally available and the chlorine at position C-3 makes it as active as cefaclor. An important structural change in the development of second generation cephalosporins was the introduction of an α-iminomethoxy group to the C-7 side chain. This gave an increased resistance to β-lactamases due to stereochemical blocking of the beta-lactam ring. Cefuroxime was the first cephalosporin to incorporate this side chain. Another very important group in the second generation is the aminothiazole ring to the C-3 side chain. This development drastically increased binding affinity to PBP and increased antimicrobial activity. The aminothiazole ring can be seen in the structure of cefotiam.
== Treatment == There is no cure for McLeod syndrome; the treatment is supportive depending on symptoms. Medication may assist with management of epilepsy, and cardiac and psychiatric features, although patients may respond poorly to treatment for chorea.

Oxidative stress reflects an imbalance between the systemic manifestation of reactive oxygen species and a biological system's ability to readily detoxify the reactive intermediates or to repair the resulting damage. Disturbances in the normal redox state of cells can cause toxic effects through the production of peroxides and free radicals that damage all components of the cell, including proteins, lipids, and DNA. Oxidative stress from oxidative metabolism causes base damage, as well as strand breaks in DNA. Base damage is mostly indirect and caused by the reactive oxygen species generated, e.g., O−2 (superoxide radical), OH (hydroxyl radical) and H2O2 (hydrogen peroxide). Short-term oxidative stress is required to initiate stress response processes in plants.
Together, they manage to get the increasingly aggressive U-Haul out of the house. Demon, still addicted to opioids, takes on and loses several minimum wage jobs but also reconnects with Tommy. Hanging out at the newspaper Tommy works for, Demon begins anonymously publishing a popular comic strip. Dori reveals that she is pregnant, and Demon tries to convince her to get sober for the baby. However, Demon comes home to find that Dori has miscarried, devastating them both. Dori's drug habit gets worse, culminating in a fatal overdose. Demon moves in with Maggot, both of them rattled over Emmy’s abuse and Dori’s death. Rose tells Demon and Maggot they can find Fast Forward by the waterfall at the Devil's Bathtub where Demon’s father died. On the way there, they pass Hammer Kelly, who is fixing a flat tire in the pouring rain. Heartbroken over Emmy, he has started doing drugs. He joins them as they head to seek justice, deciding to bring his rifle. Fast Forward, who is preparing for a reckless dive into the waterfall, fatally falls when he sees Hammer aiming the rifle at him. Hammer tries to save Fast Forward but drowns in the process. June sponsors a grief stricken Demon to stay at a rehab center in Knoxville. Here, Demon resumes drawing and decides to make a graphic novel about the history of the Appalachian people. Maggot has gone to juvenile prison for supplying Hammer with drugs the night he died, and Tommy has moved to Pennsylvania to marry a girlfriend he met online. Demon stays in touch with Angus, now at college in Nashville, and develops feelings for her.

The structures are supported by collagen fibres set at right angles to each other and arranged in a three-dimensional web with the ossicles and papulae in the interstices. This arrangement enables both easy flexion of the arms and the rapid onset of stiffness and rigidity required for some actions performed under stress.

=== Antimicrobial peptides === As an arthropod, S. mutilans has no acquired immune system, which leads to the development of numerous antimicrobial peptides in its innate immune system. An antimicrobial peptide named scolopendrasin VII was discovered in S. mutilans . The peptide stimulates actin polymerization; the subsequent chemotactic migration of macrophages was also stimulated by this peptide through activating ERK (extracellular signal-regulated kinases) and protein kinase B (Akt) activity. Scolopendrasin VII was also found to stimulate chemotactic migration of FPR1-transfected RBL-2H3 cells (mast cells), and directly binds to FPR1. Scolopin 1 and 2 are antimicrobial peptides identified from the venom of S. mutilans. These peptides were identified by Sephadex gel filtration and RP-high-performance liquid chromatography. Both scolopin 1 and 2 were reported to show intense antimicrobial activities, which includes both Gram-positive and Gram-negative bacteria and fungi. It was also reported to have moderate haemolytic effects on human and rabbit red blood cells. Scolopin 1 holds the potential as an antimicrobial agent. Scolopin 2 was found able to inhibit HeLa cell proliferation in cancer research; it was also found to combine with mitochondria, which allows it to regulate apoptosis in pathways in HeLa cells. Scolopin 2 is able to significantly suppress tumour growth in mice trials without causing any side effects. It was hence considered a strong candidate for HeLa cervical cancer treatment.
=== Hormone secretion === Two hormones are classically considered as being related to the posterior pituitary: oxytocin and vasopressin. These hormones are created in the hypothalamus and released in the posterior pituitary. After creation, they are stored in neurosecretory vesicles regrouped into Herring bodies before being secreted in the posterior pituitary via the bloodstream.
Some nuclei can undergo double beta decay (2β) where the charge of the nucleus changes by two units. Double beta decay is difficult to study, as it has an extremely long half-life. In nuclei for which both β decay and 2β are possible, the rarer 2β process is effectively impossible to observe. However, in nuclei where β decay is forbidden but 2β is allowed, the process can be seen and a half-life measured. Thus, 2β is usually studied only for beta stable nuclei. Like single beta decay, double beta decay does not change A; thus, at least one of the nuclides with some given A has to be stable with regard to both single and double beta decay. "Ordinary" 2β results in the emission of two electrons and two antineutrinos. If neutrinos are Majorana particles (i.e., they are their own antiparticles), then a decay known as neutrinoless double beta decay will occur. Most neutrino physicists believe that neutrinoless 2β has never been observed.
Shubich used a 0.5% acidic solution of Bismarck brown to contrastively stain mast cell granules in yellow-brown without staining other types of cells. Hematoxylin & Eosin (H&E) staining is non-effective for selective mast cell staining because hematoxylin does not bind to mast cell granule components. It can be used to counterstain cellular nuclei of mastocytes.

No. Melanotan-2 is manufactured synthetically. The naturally occurring peptide in the same family is alpha-melanocyte-stimulating hormone, which the body produces as part of normal endocrine and neural signalling.
Both are synthetic analogues derived from the same parent hormone. Reference descriptions usually characterise afamelanotide as more selective for the MC1R subtype, whereas melanotan-2 is reported to interact with a wider range of melanocortin receptors.
Early studies explored melanocortin signalling and pigmentation, which later led to informal use of the label tanning peptide. That label is not a regulatory category and does not imply approved status for any purpose.
No. It is a laboratory-made peptide, while the natural hormone is alpha-melanocyte-stimulating hormone, a longer peptide produced by the pituitary gland and by skin cells. Melanotan-2 mimics only a short active region of that hormone and contains non-natural residues such as D-phenylalanine.
