Published research on the compound remains limited. Much of the human data comes from small, early-stage studies rather than large controlled trials, and several questions about effects and variability between individuals remain open. Investigators have examined receptor activity, pigment pathways, and related physiological responses in laboratory and animal models. Findings from those models do not automatically translate to human outcomes. Reviews frequently note the scarcity of rigorous clinical evidence and call for better-characterized study material.
Because the substance circulates mainly through informal markets, verification is a recurring theme in technical discussion. Independent analyses have found that labeled content and actual content can diverge, and that purity varies between samples. Analytical laboratories use reversed-phase chromatography to separate components and mass spectrometry to confirm identity. Isotope-labeled internal standards improve quantification in complex matrices. Such methods describe what a sample contains but say nothing about its sterility, lawful status, or suitability for any use. Open questions remain about how consistently testing is applied across the supply chain.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.
Melanocytes are the pigment-producing cells of the skin, and they carry melanocortin-1 receptors on their surface. When the receptor is activated, cyclic adenosine monophosphate rises inside the cell and raises the activity of enzymes such as tyrosinase, which increases melanin output. Melanotan-2 binds melanocortin-1 receptors in vitro and in animal models, and this binding is generally described as the basis for the tanning effect. Other receptors account for different effects: melanocortin-4 receptors contribute to appetite and erectile signalling, while melanocortin-3 and melanocortin-5 receptors contribute to energy balance and exocrine function.

Regulatory treatment varies by jurisdiction and has changed over time. In several countries the peptide is handled as an unapproved prescription medicine, and import or sale for human use is restricted, while elsewhere it falls under poisons or controlled-substance schedules. Enforcement activity against online vendors has been reported in Australia, New Zealand, the United Kingdom and the United States. Scholarly writing discusses melanotan-2 chiefly as an experimental tool and as a case study in unregulated peptide supply, and its precise legal position in any given country should be checked against current national schedules.
Lyophilised melanotan-2 is comparatively robust when kept dry, cold and dark, and a desiccated powder stored at minus twenty degrees Celsius or below is generally expected to retain its chemical integrity for extended periods. In solution the peptide is far less stable, with degradation proceeding through oxidation of tryptophan and histidine residues, hydrolysis adjacent to the lactam bridge, and aggregation at higher concentrations. Repeated freeze-thaw cycling accelerates loss of the parent peak. Working aliquots are therefore prepared once, held cold, and used without letting the stock return to ambient temperature.
Structurally, Melanotan-2 retains the core recognition motif of alpha-melanocyte-stimulating hormone while adding a lactam bridge that links two side chains and constrains the molecule into a ring. This modification lowers susceptibility to enzymatic degradation. The compound acts as an agonist at melanocortin receptors, particularly subtypes associated with melanin production. Because the same receptor family influences several physiological processes, researchers note that its activity is not confined to pigmentation alone. Receptor selectivity continues to be examined in published studies.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its sequence incorporates modified residues that increase potency and extend biological activity relative to the native hormone. The compound binds receptors of the melanocortin family and is examined mainly in laboratory research. It does not occur naturally and exists only as a manufactured chemical entity produced by solid-phase synthesis.
The peptide was developed during the 1980s by researchers investigating melanocortin signalling and skin pigmentation pathways. Early work focused on analogues of alpha-melanocyte-stimulating hormone that would resist enzymatic breakdown more effectively than the parent molecule. Melanotan-2 emerged from that programme as a shortened, cyclised variant. Reports describing its synthesis and receptor activity later appeared in the scientific literature. Commercial availability grew through unregulated channels rather than through pharmaceutical approval.
Although probably unrelated to AMPD deficiency, if the person happens to have a high load of d-lactate in the blood (mostly from food and colonic fermentation), the precipitate, the lactate loss and the magnesium loss may occur even before l-lactate (mostly from muscles) reaches its renal re-absorption threshold. This happens because l-lactate and d-lactate compete with each other for renal re-absorption, and because d-lactate has a significantly lower renal re-absorption threshold, <1 mmol/L. In order to keep the excreted metabolites in solution, the kidney also has to excrete water. This is in contrast to complete oxidation of lactic acid, which would actually yield metabolic water for the body. This may lead to onset of acute thirst some tens of minutes into exercise in this state, if the water balance in the body was neutral initially. If the muscle load is small, lactate is mostly recycled back into glucose or burned by other cells in the body. However the newly generated glucose is made available to all cells in the body, not just to muscle cells. The ability of the body to assimilate lactate may also be diminished, if working muscle cells cannot take up glucose from blood, due to myophosphorylase maintaining a higher concentration of it inside loaded cells, and if liver has already filled its glycogen stores up to capacity. So, ultimately, in this state, working muscle cells are destined to lose all glycogen. AMP breakdown to adenosine in this state is minor, because the pool of AMP is kept small by the vigorous regulatory action of myophosphorylase.
It has been recorded growing under Pinus and Tsuga in Sagarmatha National Park in Nepal, as well as in the Indian forests of Arunachal Pradesh. In West Asia, the species has been reported from the northwest forests of Iran.

=== Recent Interests on PIDD1 === Over the past few years, biallelic pathogenic variants in the CRADD gene have been strongly associated with a rare neurodevelopmental disorder (MRT34; MIM 614499), known as the "thin" lissencephaly (TLIS) variant. This disorder is characterized by pachygyria, primarily affecting the anterior regions of the brain, and is accompanied by megalencephaly, epilepsy, and intellectual disability (ID). This discovery has drawn increased attention to the PIDDosome complex, revealing a wider array of biological functions beyond its conventional role in apoptosis triggered by DNA damage. In this regard, PIDD1, a CRADD-interacting protein, functions as a sensor that monitors centrosome numbers and plays a vital role in controlling cellular differentiation during key processes such as organogenesis and tissue regeneration. Recently, four homozygous variants in the PIDD1 gene have been identified among 11 individuals from five separate families, all of whom present with nonsyndromic intellectual disability. Nevertheless, comprehensive clinical and neuroimaging data for these cases are limited.
== Scoville organoleptic test == In the Scoville organoleptic test, an exact weight of dried pepper is dissolved in alcohol to extract the heat components (capsaicinoids), then diluted in a solution of sugar water. Decreasing concentrations of the extracted capsaicinoids are given to a panel of five trained tasters, until a majority (at least three) can no longer detect the heat in a dilution. The heat level is based on this dilution, rated in multiples of 100 SHU. Another source using subjective assessment stated, "Conventional methods used in determining the level of pungency or capsaicin concentration are using a panel of tasters (Scoville organoleptic test method). ... Pepper pungency is measured in Scoville heat units (SHU). This measurement is the highest dilution of a chili pepper extract at which heat can be detected by a taste panel." A weakness of the Scoville organoleptic test is its imprecision due to human subjectivity, depending on the taster's palate and number of mouth heat receptors, which vary widely among subjects. Another shortcoming is sensory fatigue; the palate is quickly desensitized to capsaicinoids after tasting a few samples within a short time period. Results vary widely (up to ± 50%) between laboratories.

The synthesis of luciferin exemplifies another strategy of isolating reaction partners, which is to take advantage of rarely-occurring, natural groups such as the 1,2-aminothiol, which appears only when a cysteine is the final N' amino acid in a protein. Their natural selectivity and relative bioorthogonality is thus valuable in developing probes specific for these tags. The above reaction occurs between a 1,2-aminothiol and a 2-cyanobenzothiazole to make luciferin, which is fluorescent. This luciferin fluorescence may then be quantified by spectrometry following a wash, and used to determine the relative presence of the molecule bearing the 1,2-aminothiol. If the quantification of non-1,2-aminothiol-bearing protein is desired, the protein of interest may be cleaved to yield a fragment with a N' Cys that is vulnerable to the 2-CBT.
In the 1950s, the de-Stalinisation of the Soviet Union was ideological bad news for the People's Republic of China because Soviet and Russian interpretations and applications of Leninism and orthodox Marxism contradicted the Sinified Marxism–Leninism of Mao Zedong—his Chinese adaptations of Stalinist interpretation and praxis for establishing socialism in China. To realise that leap of Marxist faith in the development of Chinese socialism, the Chinese Communist Party developed Maoism as the official state ideology. As the specifically Chinese development of Marxism–Leninism, Maoism illuminated the cultural differences between the European-Russian and the Asian-Chinese interpretations and practical applications of Marxism–Leninism in each country. The political differences then provoked geopolitical, ideological and nationalist tensions, which derived from the different stages of development, between the urban society of the industrialised Soviet Union and the agricultural society of the pre-industrial China. The theory versus praxis arguments escalated to theoretic disputes about Marxist–Leninist revisionism and provoked the Sino-Soviet split (1956–1966) and the two countries broke their international relations (diplomatic, political, cultural and economic). China's Great Leap Forward, an idealistic massive reform project, resulted in an estimated 15 to 55 million deaths between 1959 and 1961, mostly from starvation.
Retinol is made industrially via total synthesis using either a method developed by BASF or a Grignard reaction utilized by Hoffman-La Roche. The two major suppliers, DSM and BASF, are believed to use total synthesis. The world market for synthetic retinol is primarily for animal feed, leaving approximately 13% for a combination of food, prescription medication, and dietary supplement use. The first industrialized synthesis of retinol was achieved by the company Hoffmann-La Roche in 1947. In the following decades, eight other companies developed their own processes. β-Ionone, synthesized from acetone, is the essential starting point for all industrial syntheses. Each process involves elongating the unsaturated carbon chain. Pure retinol is extremely sensitive to oxidization and is prepared and transported at low temperatures and oxygen-free atmospheres. When prepared as a dietary supplement or food additive, retinol is stabilized as the ester derivatives retinyl acetate or retinyl palmitate. Before 1999, three companies, Roche, BASF, and Rhone-Poulenc controlled 96% of global vitamin A sales. In 2001, the European Commission imposed total fines of 855.22 Euros on these and five other companies for their participation in eight distinct market-sharing and price-fixing cartels that dated back to 1989. Roche sold its vitamin division to DSM in 2003. DSM and BASF have the major share of industrial production.
==== Lip biopsy ==== A lip/salivary gland biopsy takes a tissue sample that can reveal lymphocytes clustered around salivary glands, and damage to these glands from inflammation. This test involves removing a tissue sample from a person's inner lip/salivary gland and examining it under a microscope. On such biopsies, the single most important test result in the diagnosis of the oral component of Sjögren’s is likely the focus score, which is the number of mononuclear cell infiltrates containing at least 50 inflammatory cells in a 4 mm2 glandular section. The Chisholm-Mason grades are also widely used for salivary gland biopsies (see table).
=== Raynaud's phenomenon === Raynaud's phenomenon is frequently the first manifestation of CREST/lcSSc, preceding other symptoms by years. Stress and cold temperature induce an exaggerated vasoconstriction of the small arteries, arterioles, and thermoregulatory vessels of the skin of the digits. Clinically this manifests as a white-blue-red transition in skin color. Underlying this transition is pallor and cyanosis of the digits, followed by a reactive hyperemia as they rewarm. When extreme and frequent, this phenomenon can lead to digital ulcerations, gangrene, or amputation. Ulceration can predispose to chronic infections of the involved site.

While it is constitutionally a multi-party democracy where free elections are regularly held, it functions as a de facto one-party state, with the People's Action Party (PAP) maintaining continuous political dominance since 1959. The PAP's longstanding control has resulted in limited political pluralism and a highly centralised governance structure over national institutions. === Treatment === Consumption of the death cap is a medical emergency requiring hospitalization. The four main categories of therapy for poisoning are preliminary medical care, supportive measures, specific treatments, and liver transplantation. Preliminary care consists of gastric decontamination with either activated carbon or gastric lavage; due to the delay between ingestion and the first symptoms of poisoning, it is common for patients to arrive for treatment many hours after ingestion, potentially reducing the efficacy of these interventions. Supportive measures are directed towards treating the dehydration which results from fluid loss during the gastrointestinal phase of intoxication and correction of metabolic acidosis, hypoglycemia, electrolyte imbalances, and impaired coagulation. No definitive antidote is available, but some specific treatments have been shown to improve survivability. High-dose continuous intravenous penicillin G has been reported to be of benefit, though the exact mechanism is unknown, and trials with cephalosporins show promise. Some evidence indicates intravenous silibinin, an extract from the blessed milk thistle (Silybum marianum), may be beneficial in reducing the effects of death cap poisoning. A long-term clinical trial of intravenous silibinin began in the US in 2010. Silibinin prevents the uptake of amatoxins by liver cells, thereby protecting undamaged liver tissue; it also stimulates DNA-dependent RNA polymerases, leading to an increase in RNA synthesis.
== Expansions and sequels == Half-Life was followed by an expansion pack, Opposing Force, on November 1, 1999, developed by Gearbox Software. Players control Hazardous Environment Combat Unit (HECU) Corporal Adrian Shephard, who fights a new group of aliens, Race X, and Black Ops units after being split from his team. Gearbox developed a second expansion pack, Blue Shift, in which players control Barney Calhoun, a security guard at Black Mesa, as he attempts to escape the facility. It was developed as a bonus campaign for the Dreamcast port of Half-Life, but was released for Windows on June 12, 2001, after the port was canceled. Gearbox created a cooperative multiplayer expansion pack, Decay, exclusively for the PlayStation 2 port of Half-Life which is played through the perspectives of Gina Cross and Colette Green, two Black Mesa scientists. Half-Life 2 was released in 2004. The player controls Freeman 20 years after the Black Mesa incident in the dystopian City 17, where he joins a rebellion against an alien regime. It was followed by the episodic sequels Half-Life 2: Episode One (2006) and Half-Life 2: Episode Two (2007). After cancelling several other Half-Life projects, Valve released Half-Life: Alyx in 2020.
=== Impact on physical chemistry === The discovery of hydrogen isotopes also impacted physics in the 1940s, as nuclear magnetic resonance spectroscopy was first invented. Organic chemists now use nuclear magnetic resonance (NMR) to map protein interactions or identify small compounds, but NMR was first a passion project of physicists. All three isotopes of hydrogen were found to have magnetic properties suitable for NMR spectroscopy. The first chemist to fully express an application of NMR was George Pake, who measured gypsum (

Only a small number of early-stage human studies have been reported, and most were limited in size and duration. No large late-stage program has established a general efficacy or safety profile. The evidence base is therefore thin compared with approved medicines.
The peptide is listed as prohibited in competitive sport, so laboratories develop detection methods and monitor for its presence. Its appearance in enforcement reports reflects that classification rather than any approved athletic use. Analytical work in this area focuses on identifying the molecule and its breakdown products in biological samples.
Chromatographic separation combined with mass spectrometry identifies the peptide and estimates its amount in a sample. Testing can confirm whether the expected molecule is present and at what concentration. It cannot establish how a product was manufactured or whether it meets any purity standard.
No. It is a laboratory-made peptide, while the natural hormone is alpha-melanocyte-stimulating hormone, a longer peptide produced by the pituitary gland and by skin cells. Melanotan-2 mimics only a short active region of that hormone and contains non-natural residues such as D-phenylalanine.
